Starting material: Immature embryos

Characteristic feature: Embryogenic callus suitable for the regeneration of transgenic plants by using the Agrobacterium-mediated method or the particle bombardment technique

Culture conditions: Callus induction medium 2, 27℃ (lower limit 25℃), dark

Method for maintenance: Transfer a portion (approx. 2 mm in diameter) of growing callus to a fresh medium every 10 days. The growth of callus may be much slower under lower temperature such as 25℃. If it is the case, you would better wait for 14 days untill next subculture.
